It is naturally present in tarragon essential oil. It is insoluble in water, soluble in ethanol, ether, acetone, and easily soluble in benzene. It irritates the eyes and mucous membranes. It was originally obtained from the distillation of methyl salicylate or methoxybenzoic acid. It is now mainly prepared by reacting methylating agent dimethyl sulfate with phenol in alkaline aqueous solution. Anisole is prone to electrophilic substitution reactions on the aromatic nucleus. It condenses with formaldehyde to form a viscous oil or resin; reacts with phosphorus chloride to form p-chloroanisole and a small amount of ortho-chloro product; reacts with sulfuryl chloride to obtain 2,4,6-trichloroanisole. In addition, when anisole is heated with hydrobromic acid or hydroiodic acid, the carbon-oxygen bond breaks to form phenol and methyl halide, which is an important method for determining methoxy groups on the benzene ring.
